Treg cells from pooled spleen and lymph nodes of vTreg53 TCR transgenic mice were transferred into CD45.1 B6 mice. Six weeks later, the recipient mice were immunized subcutaneously with CFA/Fat1562, followed by a boost with IFA/Fat1562 intraperitoneally one week later. The clonotype+ Tregs from the spleen/lymph nodes and VAT of recipient mice were double sorted and processed for RNA-seq. We identified that peptide immunization is associated with TCR activation and that the VAT Treg transcriptome is largely maintained in the presence of antigen stimulation.
